چکیده

In this study, we investigated the influence of different modes magnetic mixing on effective enzyme activity aspartate ammonia-lyase from Pseudomonas fluorescens immobilized onto epoxy-functionalized nanoparticles by covalent binding (AAL-MNP). The specific AAL-MNPs in traditional shake vial method was compared to MNP-based biocatalyst two devices designed for agitation. first device agitated moving permanent magnets at opposite sides a x-axis direction (being perpendicular y-axis vial); second unsettled MNP rotating around vial. vial, substrate and move same with pattern. agitation modes, MNPs responded differently external field magnets. axial mode, formed cloud inside whereas they ring. Especially, generated small fluid flow enabling reaction mixture, leading enhanced

Structural basis for the catalytic mechanism of aspartate ammonia lyase.

Aspartate ammonia lyases (or aspartases) catalyze the reversible deamination of L-aspartate into fumarate and ammonia. The lack of crystal structures of complexes with substrate, product, or substrate analogues so far precluded determination of their precise mechanism of catalysis. Here, we report crystal structures of AspB, the aspartase from Bacillus sp. YM55-1, in an unliganded state and in ...

Luciferase and luciferin co-immobilized mesoporous silica nanoparticle materials for intracellular biocatalysis.

We report a gold nanoparticle (AuNP)-capped mesoporous silica nanoparticle (Au-MSN) platform for intracellular codelivery of an enzyme and a substrate with retention of bioactivity. As a proof-of-concept demonstration, Au-MSNs are shown to release luciferin from the interior pores of MSN upon AuNP uncapping in response to disulfide-reducing antioxidants and codeliver bioactive luciferase from t...

Yeast Phenylalanine Ammonia-lyase

I?henyialanine ammonia-lyase from the yeast Rhodotorula gluiinis was purified by salt fractionations and Sephadex chromatography. Density gradient centrifugation and Sephadex chromatography indicated its molecular weight to be about 275,000. Enzymatic deamination of several ring-substituted phenylalanine analogues and n-phenylalanine was studied. While cinnamic acid, a product of deamination, a...
